I have  purchased this  thing ,  and I'm thinking to use it with this connected to the timer.
My concern is that all the water from anus device will go  into bowel before valve will be open, or it will not because there is no air to come in from the opposite side ?

Not sure I follow.
The valve, if electric, may require an amount of pressure for it to work.
Also, these valves are one way, so air or water can only travel one way.
Not both ways.

It has to be a one way valve. 
Water bag > tube > valve > tube > anal device

I want to avoid air to be pushed from anal device into guts when valve comes into open state.  If I let water to fill the whole system when valve is open. then close the valve,  then push the device into anus, will the water from anal part and tube leak into guts ?

I'm thinkin' about period of a 15 min waiting period with valve closed, 1 min open , 1 close....  and then finished with a close state so the water could not run  back way to a bag because of a pressure from a body.

There will always be a bit of air in the system.
To do what you want, you will have to the following:
Fill the bag with your enema. Then open the valve so the enema will fill the tube and nozzle with the enema. Cork the tip of the nozzle to hold in the enema and turn off the valve.
All the air will now be gone.

Now, comes the tricky part. If you uncork the tip to insert the nozzle in your rump, your enema will leak out and will be replaced with an air bubble.

But, if you can plug the nozzle with a soft food item, Banana would work or even Peanutbutter, then the enema will not leak out. When the timer goes off, the valve will open, and the enema will pop out the plug and you'll get the full enema.
